It is for auditioning files from the File-Import dialog.
The port connections are configured in the Preferences dialog.
Ardour2: Window->Preferences (Ctrl+O).

Assignment of the click ports is in Preferences dialog, too. You can
also load any sample to be used as click there.

The click is defined by Meter and Tempo (the time-rulers at the top of
the canvas). The click itself can be en/disabled with the button right
of the clock.

Right-click on the tempo and meter bars: New Tempo, New meter.. or
change the default ones: right-click on the markers and edit them..

Ardour assumes that you start recording with the click. Assigning tempo
later to some pre-recorded master-track is not standard procedure. But
you might be able to whip something up using the Window -> Rhythm-ferret.
