I would use the line in. I would only use the insert jack if I was
actually using it as a loop, unless I knew it was OK to do otherwise.

It depends on if you like the tube pre better than a computer model of a
pre-amp. I would imagine that the answer would be yes, you do want to
use the pre-amp.
